Tool plugins have access to user clicks in the editor using MouseInput() and GetCursorInfo(). This gives you the coordinates of the cursor and you can check for clicks and drags using the EditorWindow* 'win' argument. You will, of course, need to figure out which points are involved. This could be selected points or the points within a radius of the screen click location.
There are a few Tool plugin examples in the cinema4dsdk source folder.
